E7080 is an orally active multi-targeted kinase inhibitor whose targets include vascular endothelial growth factor receptors (VEGFR), fibroblast growth factor receptor (FGFR) and platelet derived growth factor receptors (PDGFR). It has been shown to inhibit tumor angiogenesis by targeting endothelial cells. E7080 has shown promising activity as an anticancer agent with acceptable toxicity.
Given the lack of treatment options for patients with NSCLC in the third-line setting, the absence of approved agents for patients who have failed 3 or more treatment regimens, the potential activity of antiangiogenic factors, and the responses seen with E7080 in heavily pretreated patients, a Phase II study in patients with non-squamous NSCLC who have failed at least 2 prior systemic anticancer regimens is warranted.

E7080 je multikinázový inhibitor účinný při perorálním podávání, který inhibuje receptory pro vaskulární endoteliální růstový faktor (VEGFR), receptory pro fibroblastový růstový faktor (FGFR) a receptory pro růstový faktor odvozený od trombocytů (PDGFR). Bylo prokázáno, že svým působením na endoteliální buňky inhibuje angiogenezi v nádoru. E7080 prokázal slibnou účinnost jako protinádorová léčba s přijatelnou toxicitou.
Vzhledem k nedostatku léčebných možností pro pacienty s NSCLC jako třetího terapeutického režimu, k absenci schválených léků pro pacienty, u nichž selhaly 3 a více terapeutických režimů, k potenciální účinnosti antiangiogenních faktorů a k léčebné odpovědi zjištěné u E7080 u pacientů po rozsáhlé předchozí léčbě, je studie II. fáze u pacientů s NSCLC neskvamocelulární histologie, u nichž selhaly 2 předchozí režimy protinádorové léčby, jistě zcela na místě.
