This dates from a time when addresses often consisted not of house numbers and street names, but of approximate locations.

In English, 'at the sign of' often refers to the sign outside a tavern, which may give its name to a whole district, such as Elephant and Castle in London.

I assume 'at the sign of S. Luigi' is the area around a church. Carlieri is be the name of the publisher/bookseller.

Aldo Manuzio usò la sua famosa marca (l’ancora con il delfino) soltanto a partire dal 1502, cioè otto anni dopo l’inizio della sua attività. L’importanza pratica dell’insegna deve sempre essere tenuta presente. Costituiva una garanzia contro le numerose edizioni contraffatte.
