De nombreux laboratoires utilisent un test de quantification des anticorps anti- ADN double brin par ELISA (dosage immuno-enzymatique), un test très sensible et fiable.

D’autres anticorps peuvent être recherchés comme les anticorps anti-ADN simple brin (mais moins spécifiques du LED et sont retrouvés dans d’autres maladies auto immunes) ou des anticorps anti-antigènes nucléaires solubles tels les anticorps anti-Sm (peu fréquent dans le LED mais très spécifiques) ou encore les anticorps anti-Ro/SSA et La/SSB et enfin les anticorps anti-ribonucléoprotéines.
