color development Explanation: ELISA is based on a chemiluminescent reaction catalyzed by the linked enzyme which processes the substrate to produce a detectable signal in the form of color development (usually yellow). This coloration is what's being detected, but since it's an enzymatic reaction, it is time- and temperature-dependent. So if the reaction is run at a lower temperature, color development will take longer, but if the reaction is allowed to run for too long, it will saturate the signal and affect quantification. |
